目的对比研究胶原水凝胶和基质胶作为骨组织工程支架表面涂层材料的效果。方法本实验借助原代成骨细胞,通过对碱性磷酸酶(ALP)活性、组织学染色及成骨特异性mRNA水平的分析,研究胶原水凝胶和基质胶对成骨分化的影响。结果胶原组与基质胶组细胞活力以及增殖能力均增强,并上调成骨相关基因及蛋白,包括碱性磷酸酶(ALP)、骨钙素(OCN)、骨桥蛋白(OPN)和Ⅰ型胶原蛋白(COL1A1),尤其是胶原水凝胶涂层对细胞生长和成骨分化的促进作用明显大于基质胶涂层。结论胶原水凝胶涂层比基质胶涂层更有利于成骨分化,更适合作为细胞支架,推荐其作为骨组织工程支架材料的常用表面涂层材料。
ObjectiveTo compare the modification effect between collagen hydrogel and matrigel as surface coating of scaffold materials for bone tissue engineering. MethodsThe alkaline phosphatase (ALP) activity, hitological staining and mRNA level of osteogenic specific genes were analyzed in the primary osteoblasts, then the effects of collagen hydrogel and matrigel on osteogenic differentiation were investigated. ResultsIn the collagen group and matrigel group, the cell vitality and proliferative capacity increased, the osteogenic related genes and proteins including ALP, osteocalcin, osteopontin and alpha1 type I collagen were up-regulated. Coating with the collagen hydrogel promotion was more effective in promoting the cell growth and osteogenic differentiation compared to coating with the matrigel. ConclusionCoating with the collagen hydrogel is more beneficial to osteogenic differentiation and is more suitable for improvement of cell scaffold compared to coating with matrigel, and is recommended as as surface coating of scaffold materials for bone tissue engineering.
